St. Ambrose Episcopal Church was established in 1925 at 125 West 130th Street, now home to Beulah Baptist Church. Within a few years, St. Ambrose moved to 160 West 129th Street (now the Faith Mission Christian Fellowship). In 1936, the congregation acquired their present church, a neo-Gothic building designed by James W. Pirsson and built from 1873-75 for the Presbyterian Church of the Puritans (originally known as the Second Presbyterian Church of Harlem).

The organ in St. Ambrose Church was built in 1961 by Austin Organs of Hartford, Conn. Austin installed the pipes and mechanism in the west transept gallery, and placed the three-manual console in the opposite gallery.

The original organ in the present building was built in 1875 by George Jardine & Son for the Church of the Puritans. That same year the Church of the Puritans suffered "financial embarrassment" and the organ was removed. It is believed that this organ was moved to St. Cecilia R.C. Church on East 106th Street in Harlem, where it was extant in 2010. Some pipes on the St. Cecilia organ are marked "Church of the Puritans." Following is the specification of the organ now in St. Cecilia Church.
