Warren Street Methodist Episcopal Church began as a mission in 1852. The first building was located on the corner of Court and Butler Streets. From 1856-1860, the mission met in a hall at the corner of Court and Sackett Streets. In 1860, a Gothic structure was erected at 303 Warren Street, between Smith and Court Streets. Constructed of brick with brownstone trimming, the new church measured 22 by 87 feet and provided space for 700 Sunday-school scholars in the basement, and about 1,200 in the main auditorium on the second floor. Dedicated on June 24, 1860, the new church cost about $22,000 for the building and furnishings.

At an unknown time, the congregation moved into the present modern building at 450 Warren Street.

In the early 1980s, the Cuyler Presbyterian Church merged into Warren Street United Methodist, and the consolidated congregation became the Cuyler-Warren Street United Methodist Church.
